Confocal Microscopy
3D Timelapse Imaging of Cultured Cells
Laser confocal optical sections are recorded as type
of data set that can be manipulated with software to present many views
including 3D projection effects. The confocal technique allows the light
to be collected from discrete planes while out of focus light is rejected
by a pinhole. Although the fluorescence emanating from the focal plane at each scan
point is vanishingly small its signal is amplified by a photo multiplier tube (PMT).
